レーシング・スポーツの最高峰、ピストン・カップに出場する若きレーサー、ライトニング・マックィーン。ルーキーながら、その天才的素質によって、彼は既に多くのファンを獲得していた。狙うはピストン・カップ史上初となる新人チャンピオン。しかし、決勝レースが開催されるカリフォルニアに向かう途中、思わぬトラブルに巻き込まれ、ルート66沿いにある“地図から消えた古い町、ラジエーター・スプリングス”に迷い込んでしまう。早くレース場に到着しなければならないマックィーンだが、自らが滅茶苦茶にした道路の補修工事を命ぜられ、しばらくその町に留まることになってしまうのだった。しかし、ラジエーター・スプリングスで過ごすうちに、マックィーンは徐々に住民たちに親しみを感じ始めていた。それは、自分だけが勝つことしか考えなかった独りぼっちの彼が知らなかった、《家族のような温かい関係》だった。だが、遂にレースの日がやって来た。彼は決勝レースに間に合うのか！？仲間たちとの運命は！？
