　　清雍正以來，樂家老店同仁堂被欽定為“禦藥供奉”，至樂家第八代已聲名大震。日本浪人小野勾結在樂家長大的曹化倫，蓄謀奪走秘方藥譜，行兇縱火，燒燬樂家細料藥庫。又勾結太監扣押禦藥銀子，使老店雪上加霜，內外交困。為挽救同仁堂的命運，老東家樂闊海毅然隔代傳印給長孫樂宏達。

　　樂宏達在妻子涂瑞珍的幫助下，奪回禦藥供奉，拜得到光緒皇帝的賜匾。曹化倫設計使同仁堂專供禦藥，不得為民間治病，老東家為助孫子親自出馬，一藥回天，西太后終於開禁。

　　樂家收留的女伶叢玉珠，為營救暗戀的樂宏達，捨身嫁給曹化倫。小野率日本兵闖入樂家搜取密方，樂闊海以毒酒相酬，與之同歸於盡。甲午戰爭，樂宏達趕制十萬包“諸葛行軍散”，贈送將士，卻被藥材商抬價，欠正鉅款，在無錢還債的情況下，憤而變賣祖宅。藥材商得知贈藥原因，慨然放棄債權，老店又一次絕處逢生……
