蘭貞很快就長大成人，但她波瀾起伏的一生才剛剛開始。政勢日益衰敗。慧和門外一皮鞋匠成為貫通所有人物的媒介，趙光祖等主張道學政治的士林派被擠出朝廷，蘭貞從一個幸福的頂端跌落到谷底，無情地被踐踏。為了實現自己的野心，蘭貞放棄當藝妓，嫁入尹家，成為尹元衡的妾室。蘭貞與尹元衡的姐姐，當今的文定皇后攜手，將敬嬪、金安老等政治道路上的對手逐一除掉。
中宗駕崩，明宗即位，文定王后終於如願以償掌握了國家政權。隨著一代女傑文定皇后和貞敬夫人蘭貞一手遮天的時代到來，鄭蘭貞為了成為尹元衡的正室夫人，毒死了尹元衡的原配夫人金氏。中間穿插蘭貞與吉尚，文定王后與僧人普雨的浪漫愛情。經過二十多年的專政，文定皇后逝世，女人天下也隨之落下帷幕。
