三十年前，在設有美國海軍調查站的鯊齒島上，一條變種的大白鯊肆虐後逃往大海，消失得無影無蹤。現在海洋生物學家查斯博士，跟他的前妻阿曼達及兒子麥斯，同時在鯊齒島上工作，沒想到非比尋常的事情正等待著他們。
在一次的海上行程中，三人被懷疑是大白鯊類的物體襲擊，令船隻受到嚴重破壞，三人幸保性命！他們相信襲擊自己的並非普通的鯊魚，可是除了酗酒的老伯狼人之外，這種說法完全無人相信。三人心有不甘，終於發現一神秘山洞，並找到了一隻鯊魚般的兩腳怪物！
查斯與阿曼達幾經掙扎才得以逃脫，決意要找出真相。始揭發一個重大的秘密：怪物原是被注入人類基因的大白鯊稱為海豚科食人鮫，而狼人更與事件有直接關係。查斯與阿曼達決定放下偏見，一同對付那嗜血成狂的海洋大怪物。
Thirty years after a strange shark hybrid escapes out of sea from an island naval research station, the marine biologist Simon Chase, his ex-wife Amanda and their teenage son Max are working in that same facility.
They then experience shark-like attacks against their boat and on the island but the only one who’ll believe their story is an unbalanced drunk called Werewolf. Together they explore the old naval buildings and discover a hidden cavern where they encounter a shark-like creature that stands on two legs!
Barely managing to escape its deadly jaws, Amanda and Simon dig up the old research notes and make a horrifying discovery: the creature is made from human DNA and Werewolf is directly implicated! The two divorced marine biologists join forces to stop the marauding, bloodthirsty sea monster from continuing its rampage on land.
